Research Summary

Mass spectrometry is a useful tool to identify and quantify biological molecules in complex mixtures. By using isotopic-labeling techniques (e.g., iTRAQ) coupled with tandem LC/MS the bacterial proteome can be analyzed and compared under a variety of growth and environmental conditions. The relative protein levels present in the bacteria prior to and after induction of the amorphadiene pathway yield insight into the mechanisms E. coli uses to respond to the drain on resources and important metabolite pools. A thorough understanding of the cellular processes leading to these deleterious effects will permit modification of the production system (e.g., host, media, growth conditions) to minimize or eliminate the harmful components.
